Because there's no diskette in it?
In all seriousness, what kind of drive is it? What's the OS?

what diskette?
Floppy's are dead for a long time, time 2 move on.
Its a NTFS partition in Windows 7 (64bit). it must be disabled in s/w because everybody is still treating B: as for floppy (god knows why?)

Believe it or not, floppy drives are still being used.
As far as your B:Drive, you might need to change it to another drive letter. Is it a data only drive?

A: and B: are reserved in Windows (and BIOS) for floppy drives. If you have a drive mapped there, go into safe mode and change it. First, go into the BIOS and check to see if anything is assigned there.

As A: and B: are conventionally FDDs on Windows (until Win7 apparently), this is laziness on Serato's behalf.
If you have your hard drive's boot partition as A: and your Data partition as B:, I'd rethink that...

Change it. Keep your system drive as C and data drive as anything from D to Z completely your call. Unless you have any specific reason for it keep all drives in this order. That way you wont have any further problems with lazy code :P

BTW, I've confirmed that on Win7, a storage unit can be mapped to ANY drive letter. That's NEW and will cause compatibilty issues (particularly with legacy conventions) like this one if users continue to map storage to letter BEFORE C:...

to be honest I don't care why they didnt do it.
I post a BUG and I would like to see a fix for that BUG.
Even if that is for floppy, why cant I use a FDD then?

^ I would assume that ITCH (or SSL or ANY app) would be able to locate files on any accessible drive. I say submit a help request... hopefully it's just an included library that needs to be updated during the compilation.
Of course, it'd be faster to remap your storage to a drive letter AFTER C:...
